چکیده

Mathematical modeling and computer simulation are valuable tools in unravelling the complexities underlying the morphological development of neurons. In this chapter, we consider biologically plausible mathematical models of neurite initiation, elongation, and branching. These models attempt to describe the intraand extracellular environments of a developing neuron and determine the limiting factors in neurite outgrowth. This typically involves modeling the production, degradation, and transport of one or more chemicals in one or two spatial dimensions. For example, the one-dimensional diffusion and active transport of tubulin within a neurite, or the two-dimensional diffusion gradient of a chemoattractant in the external environment surrounding a neuron grown in cell culture. Extension to three dimensions to accurately describe neuronal growth in vivo may be desirable, but is highly computationally intensive and likely analytically intractable. The numerical solution of even oneor two-dimensional models is computationally demanding and requires considerable care in specifying appropriate temporal and spatial discretization. As the neuronal morphology changes over time so must the spatial discretization representing the neuron also change. Numerical techniques that address this issue are described. Modulation of H2O2- Induced Neurite Outgrowth Impairment and Apoptosis in PC12 Cells by a 1,2,4-Triazine Derivative

Introduction: Increased oxidative stress is widely accepted to be a factor in the development and progression of Alzheimer’s disease. Triazine derivatives possess a wide range of pharmacological activities including anti-oxidative and anti-in.ammatory actions. In this study, we aimed to investigate the possible protective effect of 3-thioethyl-5,6-dimethoxyphenyl-1,2,4-triazine (TEDMT) on H2O2-...

Effects of Different Concentrations of Morphine on Staurosporine-Induced Neurite Outgrowth in Pc12 Cells

Purpose: The present study was conducted to evaluate the effect of different concentrations of morphine on staurosporine-induced neurite outgrowth in PC12 cells. Materials and Methods: PC12 cells were cultured in RPMI1640 culture medium supplemented with 0.2% BSA. Cells were divided into three groups; Ι, ΙΙ and ΙΙΙ, culture in the presence of 50, 100 and 214 nM staurosporine respectively. In ea...

Human Olfactory Ecto-mesenchymal Stem Cells Displaying Schwann-Cell-Like Phenotypes and Promoting Neurite Outgrowth in Vitro

Strategies of Schwann cell (SC) transplantation to regenerate the peripheral nerve injury involves many limitations. Stem cells can be used as alternative cell sources for differentiation into SCs. Given the high potential of neural crest-derived stem cells for the generation of multiple cell lineages, in this research, we considered whether olfactory ecto-mesenchymal stem cells (OE-MSCs) derive...

Proteolytic control of neurite outgrowth inhibitor NOGO-A by the cAMP/PKA pathway.

Protein kinase A (PKA) controls major aspects of neurite outgrowth and morphogenesis and plays an essential role in synaptic plasticity and memory. However, the molecular mechanism(s) of PKA action on neurite sprouting and activity are still unknown. Here, we report that in response to neurotrophin or cAMP stimulation the RING ligase praja2 ubiquitinates and degrades NOGO-A, a major inhibitor o...

Effects of different culture media on optimization of primary neuronal cell culture for in vitro models assay

Background: In vitro model studies are becoming increasingly popular for experimental research designs. They include isolation and expansion of cells of a particular tissue, such as the nervous tissue which contributes to understanding the underlying mechanisms in many pathologies. It enables  the scrutinization of intracellular signaling pathways responsible for cell death. OBJECTIVES: In the ...
